"ChIP assays were conducted with the use of SimpleChIP Enzymatic Chromatin IP Kit (Magnetic Beads, Cell Signaling Technology) according to the manufacturer protocol. Cross-linked chromatin prepared from 4 x 10e6 cells was immunoprecipitated overnight with antibodies against EZH2 (Cell Signaling Technology, #5246S), H3K27me3 (Cell Signaling Technology, #9733S) or TAp73 (Abcam, #ab14430). The immunocomplexes were then conjugated with ChIP-Grade Protein G Magnetic Beads (Cell Signaling Technology) for 2 hours at 4C. After 4 washes, the bead-bound DNA was eluted by gentle vortexing (1,200 rpm) for 30 minutes at 65C. Purified DNAs and the input were subjected to ChIP-seq. The TruSeq ChIP Library Preparation Kit (Illumina) was used for library preparation. Libraries were sequenced on the Illumina NovaSeq6000 platform with a standard 150-bp paired-end read protocol."
